Answer:

With the iPhone 4 and iOS 4, double-tap the Home button to bring up the Recents List. Slide your finger to the right. The screen orientation lock and iPod controls will appear. Tap the screen orientation lock to lock the screen in whatever orientation it currently appears on your iPhone. Repeat the same steps to disable the screen orientation lock.
